中文摘要芙蓉菊(CrossostePhium chinense)是我国中南地区资源非常丰富的一种药用植物,其水煎液在民间被用来防治糖尿病。本文以芙蓉菊干燥植株为原料,经过多级提取得到芙蓉菊粗多糖液,预处理后用 部分酸水解法水解成为芙蓉菊寡糖片断,之后用活性炭柱层析分离得到四种洗脱组分。应用高糖致胰岛细胞损伤和STZ致胰岛细胞氮氧自由基损伤模型对上述洗脱组 分进行生物活性测定。正常胰岛细胞分泌中空白组胰岛素含量为7.694 mIU/l;芙蓉 菊寡糖水洗脱组分的胰岛素含量为8.506 mIU/l;5%乙醇洗脱组分的胰岛素含量为 9.689 m IU/l。可见,芙蓉菊寡糖水、5%乙醇洗脱组分有促进胰岛素分泌的作用。芙蓉菊寡糖水、5%乙醇洗脱组分经凝胶色谱柱洗脱曲线检验其组分均一度良好; 经过基体辅助激光解吸一电离质谱(MALDI-TOF)验证芙蓉菊寡糖5%乙醇洗脱组分 摩尔质量是7OID。,可以推测该组分是一个四糖单元加上一个钾金属离子的复合物;其红外光谱体现了糖类物质的特征吸收峰,该组分是一种四糖物质。另外本文还探索了简单易行的工艺和技术用以制备甘露糖醛酸锡络合物,用正交 设计进行了参数优化。得到最佳反应条件为Time=1hr,pH=8,MM=0.5g/150 ml, CdCl2=1.0g/75ml,柠檬酸三钠=1.5g,此时得到络合物产率为54.6%,络合物溶解 度为24.1g/100 ml H2O,搞含量为9.2%,甘露糖醛酸有可能是一种排除福毒的新型的福络合剂。
英文摘要Crossostephium chinense is a kind of Chinese traditional herb distributed abundantly in the midsouth of China, and it's aqueous extract is widely used for treating and preventing Diabetes Mellitus. One kind of polysaccharide was isolated from the dried Chinese hevb-Crossostephium chinense by fractional preparation, and after pretreatment this polysaccharide was hydrolyzed by acid incompletely to be a complex of oligosaccharides. The activated carbon column chromatography was used to separate it and four kinds of oligosaccharide extracts were obtained by different elution liquid. Based on islet impairment model by high blood glucose, tested the level of insulin secretion promoted by these four oligosaccharide extracts. Based on STZ-induced diabetes islet impairment model by NO free radical, tested the level of NO content affected by these four oligosaccharide extracts. The blank insulin secretion of normal islets was 7.694 m IU/1, oligosaccharide aqueous extract was 8.506 m IU/ 1 and oligosaccharide 5% ethanol extract was 9.689 m IU/1. The results indicated that Crossostephium chinense oligosaccharide aqueous extract and 5% ethanol extract could promote insulin secretion. Using the gelatin chromatography, the uniformities of Crossostephium chinense oligosaccharide aqueous extract and 5% ethanol extract were tested to be good. Using the MALDI-TOF, the molecular weight of Crossostephium chinense oligosaccharide 5% ethanol extract was tested to be 701Da, this extract can be speculated to be a compound of a kind of tetrasaccharide and a potassium ion. On the other side, Crossostephium chinense oligosaccharide 5% ethanol extract can be speculated to be a tetrasaccharide, because the IR chart of this extract was proved to contain characteristic apex of tetrasaccharide. In addition, cadmium - mannuronic complex (MMCd) was successfully prepared by using easy and simple techniques and the factors were optimized orthogonally. The Five most suitable factors were time = 1 hr, pH=8, mannuronic=0.5 g/150 ml, CdCb^l.O g/75 ml, Na3C6H5O7=1.5 g. Under these conditions the productivity of complex is 54.6 %, the solubility is 24.1 g/100 ml H2O, the cadmium content is 9.2%. Mannuronic may become a new chelating cadmium agent to eliminate cadmium toxicity.
