Separation and analysis of sialyglycopeptides extracted from egg yolks

英文摘要Objective To establish the HPLC analysis method of sialylglycopeptide (SGP) , to optimize the extraction process. Methods Harnessing a Hilic hydrophilic column,a C18 reversed-phase column,and a high-performance anion-exchange column; Optimizing HPLC elution conditions to analyze sialylglycopeptide crude samples,and then compare and optimize the separation of dual-sialylglycopeptide (d-SGP) and monosialylglycopeptide (m-SGP) using three different HPLC columns; The separation efficiency was evaluated based on peak shape, retention time interval,peak symmetry,and separation resolution. Then single factor optimization method was adopted to evaluate the extraction rate of d-SGP. The extraction times,amount of ethanol,extration duration were selected as the optimization factors to optimize the extraction process. Results Under optimal HPLC conditions, the separation of d-SGP and m-SGP indicated better resolution with the Hilic column and the anion-exchange column than with the C18 column. The optimal extraction process is using 60% ethyl alcohol as the extracting solution,extracting 60 min each time for 3 times at a ratio of egg yolk powder: extracting solution (1: 5) . Conclusion In the study of the separation of sialylglycopeptides,the Hilic chromatographic column is the best for the separation of SGP samples. The optimized extraction process is feasible.
