中文摘要 | oral cancer is the eighth most common cancer worldwide with an overall 5-year survival rate of less than 50%. precise differentiation of benign and malignant tumors plays key roles in proper and timely selection of the therapeutic treatment. this study will pay more attention to screening valuable markers by comparing a more complete metabolite profiles in the relevant biofluids (plasma, urine and saliva) from the control, benign and malignant oral tumor patients. in the current study, plasma, urine and saliva metabolic profiles for three groups consisting of malignant oral tumor patients, benign oral tumor patients and healthy controls were performed using ultra performance liquid chromatography coupled with quadrupole/time-of-flight mass spectrometry and gas chromatography coupled with mass spectrometry, respectively. after peak alignment, 964 metabolic features had been detected from three metabolic profiles including 409 from plasma, 448 from urine and 107 from saliva, respectively. utilizing multivariate chemometric analysis, excellent discriminations were obtained among the three groups based on their plasma, urine and saliva metabolic profiles, respectively. over twenty discriminatory metabolites from plasma, urine and saliva were identified. the identified metabolites were discussed in the context of their involved pathways and biological activities. these results indicated malignant oral tumor patients show altered energy metabolism, a disorder lipolysis and a distorted krebs cycle compared with benign oral tumor patients and healthy controls. in addition, plasma, urine and saliva metabolism are intimately correlated in human body. our results implied that a more complete picture of the metabolic pathways in oral cancer can be achieved by integrating plasma, urine and saliva profiles of the same patients and controls simultaneously. it will helpful for further underlying mechanisms of oral cancer. |
