中文摘要新疆沙冬青(Ammopiptanthus nanus(M.Pop.)Cheng f.)起源于古地中海热带地区,是中国温带荒漠区系中仅有的2种常绿阔叶孑遗灌木之一,可在冬季-20℃~-30℃的气候条件下保持常绿。用考马斯亮蓝法、茚三酮比色法和苯酚法,对吐鲁番沙漠植物园内引种栽培的新疆沙冬青的叶片中可溶性糖、可溶性蛋白和游离脯氨酸的季节变化过程进行了测定,并分析讨论了这3种渗透调节物质的变化与主要气象因子的关系;用DEAE-纤维素DE-52、丙烯葡聚糖S300柱层析和SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)分离和纯化新疆沙冬青叶片热稳定蛋白和抗冻蛋白(antifreeze proteins);用差示扫描量热法(DSC)测定抗冻蛋白的热滞活性(thermal hysteresis activity)。结果显示: 1.(1)可溶性糖与气温呈极显著负相关(P<0.01),曲线拟合为三次曲线(R2=0.9582,p<0.01),在-3.8℃~23.8℃之间,可容性糖对新疆沙冬青的低温适应性起渗透调节作用;(2)可溶性糖与光照呈极显著的负相关(P<0.01),曲线拟合为三次曲线(R2=0.8776,p<0.01),在90.2~322.5μmol?m-2?s-1之间,随光照强度的降低而不断积累;(3)可溶性糖与空气相对湿度呈极显著正相关(P<0.01),曲线拟合为二次曲线(R2=0.6505,p<0.05),随空气相对湿度的升高而不断积累;(4)可溶性蛋白与空气相对湿度呈显著正相关(P<0.05),曲线拟合为三次曲线(R2=0.6777,p<0.05),在18.7%~51.7%之间,随空气相对湿度的升高而不断积累;(5)游离脯氨酸与空气相对湿度呈显著正相关(P<0.05),曲线拟合为双曲线(R2=0.565,p<0.01),随空气相对湿度的升高而不断积累。 2.冬季新疆沙冬青叶片抗冻蛋白(AFPs)的热滞活性(THA),在20mg/mL时约为0.46℃,分子量约为119.24kD,表明新疆沙冬青生物体内存在抗冻活性物质—抗冻蛋白。 3.在自然分布区和吐鲁番沙漠植物园分布区的新疆沙冬青冬季叶片中分离纯化出三个热稳定蛋白,分子量分别为43.8kD、16.8kD、14.8kD。表明新疆沙冬青叶片中具有热稳定蛋白。上述研究结果和结论揭示出新疆沙冬青通过抗冻蛋白、热稳定蛋白和渗透调节物质中的可溶性糖共同调节方式来适应低温环境,对植物低温复杂的适应对策和机理研究提供有意义的理论和参考。Ammopiptanthus nanus dates from Tethy tropic area, and is one of only two evergreen broad- leaved relict shrubs in temperate hungriness flora in China. It can keep evergreen low temperature below -20℃~-30℃ in winter. The soluble sugar, soluble protein and free proline in leaves of Ammopiptanthus nanus were determined with Coomassie Breiliant Blue method, Ninhydrin Colorimetric method and Phenol method, and the relation between seasonal regular variations and main environmental factors were analysed. The antifreeze proteins and heat-stable proteins were isolated and purified with DEAE Cellulose 52, Sephacryl 300 and SDS-PAGE, and the thermal hysteresis activity (THA) of AFPs were measured using a differential scanning calorimeter (DSC). The results showed : 1.(1) soluble sugar negatively significantly correlated with air temperature (P<0.01), and curve fit accorded with cubic(R2=0.9582,p<0.01). During -3.8℃~23.8℃, soluble sugar played a role in Ammopiptanthus nanus’ osmotic adjustment under low temperature;(2) soluble sugar negatively significantly correlated with sunlight (P<0.01), and curve fit accorded with cubic(R2=0.8776,p<0.01). During 90.2~322.5μmol?m-2?s-1, soluble sugar increased while intensity of sunlight lowed;(3) soluble sugar positively significantly correlated with air temperature (P<0.01), and curve fit accorded with quadratic (R2= 0.6505,p<0.05), soluble sugar increased while air temperature raised;(4) soluble protein positively significantly correlated with air humidity(P<0.05) ,and curve fit accorded with cubic(R2=0.6777,p<0.05). During 18.7%~51.7%, soluble protein increased while air temperature raised; (5) free proline positively significantly correlated with air humidity (P<0.05) ,and curve fit accorded with inverse(R2=0.565,p<0.01), free proline increased while air temperature raised. 2. The molecular weight of AFPs was 119.24 kD by measurement. The thermal hysteresis activity (THA) of AFPs was 0.46℃ at 20 mg/mL. The above results showed there was AFPs in Ammopiptanthus nanus. 3. Three heat-stable proteins from Turpan Eremophytes Botanic Garden and the natural habitat of Ammopiptanthus nanus were isolated and purified, their molecular weight were 43.8kD, 16.8kD,14.8kD. The result showed there were heat-stable proteins in Ammopiptanthus nanus. The above results and conclusions showed that Ammopiptanthus nanus adapted low environm- ent by corporate adjustment of AFPs, heat-stable proteins and soluble sugar in osmolytes. The study provided significative theory reference for adaptive countermeasures and mechanism in plant under low conditions.
