Cloning and characterization of a drought-inducible MYB gene from Boea crassifolia

英文摘要A drought-inducible MYB gene, designated BcMYBI (GenBank accession no. AY278440), was isolated from Boea crassifolia (Hemsley) by RT-PCR and reverse northern blot screen. The full-length cDNA of BcMYBI consists of 1167 nucleotides and has an open reading frame of 1023 bp, encoding a typical R2R3-MYB transcription factor belonging to the subgroup I I of the R2R3-MYB protein family. RNA gel blot analysis showed that BcMYBI was strongly induced by drought stress, and also responded to PEG, high salinity and low temperature to some extent. However, BcMYBI transcripts did not or just slightly accumulated upon treatments with exogenous abscisic acid (ABA), methyl jasmonate (MeJa) and salicylic acid (SA). These results indicate that BcMYBI might be involved in the regulation of gene expression in response to dehydration stress through an ABA-independent pathway, whereas it seems not to be a regulatory component in wounding signaling. In addition, the 1007 bp upstream regulatory region of BcMYBI was obtained by PCR-based chromosome walking, and sequence analysis of the region predicted some putative cis-elements related to abiotic stress response. Chimeric beta-glucuronidase gene (GUS) under the control of the BcMYBI promoter was constructed and transformed into tobacco (W38). And the assays of GUS activity confirmed that BcMYBI promoter could respond to drought stress to some extent in transgenic tobaccos. (C) 2004 Elsevier Ireland Ltd.
