中文摘要 | Comprehensive analysis of whole proteomes is an extraordinary challenge because of the complexity and wide dynamic range of proteins. The success of proteome analysis is highly dependent on the quality of sample prefractionation prior to the identification by MS/MS. In our recent work, preparative microscale solution isoelectric focusing (PMSIEF) was used to achieve narrow-band fractionation of proteins from Escherichia coli (E. coli). Proteins in various pH related fractions were further separated using coupled long reversed phase high-performance liquid chromatography (RP-HPLC) columns and identified by MS/MS. |
