Involvement of N-mediated defense in oligochitosan inducing resistance to tobacco mosaic virus

中文摘要oligochitosanwas applied by spraying it on tobacco leaves for inhibition of tobacco mosaic virus (tmv). the maximum inhibition of tmv by oligochitosan was observed when inoculation occurred at 24 h after spraying 50 g/ml oligochitosan (zhao et al., 2006). but the mechanism is unknown. the tobacco n gene, a member of the toll-interleukin 1 homology region/nucleotide binding site/leucine-rich repeat (tir-nbs-lrr) class of resistance (r) gene, confers resistance to tobacco mosaic virus (tmv) (liu et al., 2004). in this report, we investigated the roles of some genes involved in n signaling, after oligchitosan treating tobacco plants. we used rt-pcr to identify changes of these known defense genes. it was found oligochitosan induced sgt1 and eds1 gene expression, any of which is required for n function. a mitogen-activated protein kinase (mapk) oipk, or an mapk kinase (mapkk) ntmek2, was up-regulated by oligochitosan. onemember of wrky transcription factors, tizz, which is concerned with n-mediated lesion forming was also induced by oligochitosan. silencing of the oipk gene attenuated oligochitosan-inducing resistance to tmv (feng et al., 2006). we also found silencing oipk altered oligochitosan-triggering genes expression, and further silenced tizz. our results suggest that oligochitosan inducing tobacco defense is involved with n-mediated resistance.
