Improved concentration technique for purified preparative fractions via solid phase extraction using oasis HLB as sorbent

中文摘要the concentration technique via solid phase extraction (spe) is a unique alternative to concentrating fractions from a preparative hplc purification [1]. comparing to direct fraction solvent evaporation, this technique has obvious advantages: increased throughput and operation at room temperature. however, five times dilution with aqueous solvent is required because collection directly onto spe cartridges offers the potential for sample loss via break through [1], which will increases greatly the volume of the fraction and limit the throughput of loading sample. so a spe sorbent with much stronger retention than c18 is necessary to load sample without dilution or with less dilution. oasis hlb sorbent has been reported with good retention for both polar and non-polar compounds [2]. we compared the curve of lnκ-acetonitrile% for five various polar compounds as measured with c18 and hlb using 5cm long column to obtain experimental data. as shown in fig. 1, the retention curves of hlb was over that of c18 for each of these compounds, which indicated that the retention of the five compounds on hlb is stronger than that on c18 under the same acetonitrile%. the data showed that the proportion of dilution is much less than five. for example, only 1.7 times dilution is needed for goitrin, a highly polar alkaloid and 1.5 time dilution is needed for quercetin, a non-polar flavonoid, by using hlb sorbent. it is concluded that the concentration by spe using hlb as sorbent reduce greatly the volume of the fraction and improve the throughput of loading sample. the concentration technique via spe using hlb sorbent instead of c18 sorbent will be more efficient for the preparation of natural products such as flavonoids and alkaloids, whether polar or non-polar. references [1] stevens, j., crawford, m., robinson, g., roenneburg, l. 2007. automated post-collection concentration for purified preparative fraction via solid phase extraction. j. chromatogr. a 1142, 81-83.
