中文摘要spicules formation in sponge cell culture in vitro xupeng cao*, xiaoying zhang, wei zhang marine bioproducts engineering group, biotechnology division, dalian institute of chemical physics, chinese academy of sciences, dalian, 116023, china silicon is the second abundant element in the earth’s crust and silica is one of the most important materials in industry. novel ways for synthesis and application of silica are highly demanded. during millions years of evolution, some organisms develop the ability to synthesize and utilize the silica. as the skeleton structure, sponge spicules are made of silica in the sea water under mild conditions. due to their special microstructures, shape diversity and excellent optical and mechanical properties, sponges spicules have potential applications as advanced silica materials in industry and for medical treatments. the study on the spicules formation mechanism is underway. a protein named “silicatein” was identified to catalyze silicic acid polymerization to form silica. progresses have been made in mimicking silicatein to produce shape-defined silica. four cdna sequences of subunits of silicatein from three sponge spices have been reported. in our work, sponge-cell-culture technique was developed to study the in vitro formation of spicules in a marine sponge, hymeniacidon perleve collected from china yellow sea. in cell culture system we observed the formation of spicules and provide experimental evidences for relationship between the growth of spicules and the secretion of collagen. the spicules formed in cell culture system are similar to the ones formed in functional sponges. partial cdna sequences of the silicatein subunits from hymeniacidon perleve hymeniacidon perleve were obtained. further studies will be discussed. acknowledgements w. zhang wishes to thank the financial support from the chinese academy of sciences under the “100 talent project”, “innovation fund” from dalian institute of chemical physics, and “hi-tech research and development program of china” (2001aa620404), and the european commission (project: silicon biotechnology).
