磷脂酶Dα在拟南芥低温驯化过程中的作用途径分析

英文摘要本研究对低温驯化和未经低温驯化的磷脂酶α基因敲除突变体与野生型拟南芥植株进行冻害胁迫处理后,通过观测植株表型和膜离子渗漏率的变化,鉴定两种基因型植株的抗冻性.结果发现,低温驯化后,PLDα敲除突变体的抗冻性明显低于野生型,表明PLDα参与植物的低温驯化过程.对其作用途径进行分析发现,PLDα不参与低温驯化过程中ABA信号转导作用,也没有参与SOD、CAT和POD等3种抗氧化酶活性的调节,但是参与低温驯化过程中膜稳定性调节和渗透调节过程.; Cold-acclimated and non-acclimated PLDα knockout and wild-type Arabidopsis thaliana plants were subjected to freezing treatment, and then changes in phenotype and membrane ion leakage were investigated to detect plant freezing tolerance of the two genotypes. The results demonstrated a lower freezing tolerance in PLDα knockout than wild-type plants when subjected to freezing stress after cold acclimation, indicating that PLDα gene is involved in the process of cold acclimation. The pathways through which PLDα acts in cold acclimation were then analyzed. PLDα was proved to be not involved in the pathway of ABA signaling under cold stress, nor involved in the regulation pathway of anti-oxidative enzymes, such as SOD, CAT, and POD. However, PLDα was found to mediate the regulation pathway of membrane stability and osmotic adjustment.
