英文摘要目的:研究SAHA对多发性骨髓瘤细胞株U266细胞增殖和凋亡的影响,观察U266细胞中乙酰化的组蛋白Ras/Raf/Mek/Erk和scf、IL-1β因子表达的变化,探讨其抗多发性骨髓瘤细胞的分子生物学机制。 方法:采用胎盘蓝计数、四氮哇蓝(MTT)比色法检测U266细胞增殖;AllllexinV和PI染色后应用流式细胞仪检测细胞凋亡率;Western-blot方法检测信号转导通路Ras/Raf/Mek/Erk的相关蛋白表达水平的改变,ELISA法检测SCF、IL-1β因子表达的变化。 结果:SAHA可明显抑制U266细胞增殖,且具有时间剂量依赖性(P; objective:To study the effects of SAHA on multiple myeloma cell line U266 eells in vitro and the underlying mechanism of SAHA in the treatment of MM were investigated.
