英文摘要	摘要 目的 应用诱导培养技术从人胶质瘤细胞系U87中诱导培养胶质瘤干细胞，分析其生物学活性。研究在胶质瘤干细胞中鞘氨醇激酶与烷化剂敏感性关系。应用鞘氨醇抑制剂诱导肿瘤干细胞分化增加烷化剂抗肿瘤效果，确定鞘氨醇激酶为胶质瘤治疗的有效靶点。 方法 通过诱导培养技术从人胶质瘤细胞系U87当中诱导培养胶质瘤干细胞。利用免疫荧光技术检测胶质瘤干细胞球CD133和巢蛋白（Nestin）。应用MTT试验检测胶质瘤干细胞和普通胶质瘤细胞对烷化剂药物尼莫司丁（ACNU）的敏感性。应用定量PCR技术比较胶质瘤干细胞和普通胶质瘤细胞的鞘氨醇激酶1基因(SPHK1)表达差异。应用MTT试验和流式细胞仪检测在胶...; Abstract 1． Objective Induction of the culture from the human glioma cell line U87 glioma cells in the induced culture to analyze the biological activity.
