| 英文摘要 | 本文对采集自太平洋深海2000m～5000m的沉积物样品进行了分子生态学调查，研究了样品中微生物多样性及群落结构并对其与环境的关系进行了分析。针对深海环境的特殊性，本文建立了一套能有效地提取沉积物中微量DNA的方法。该法采用化学裂解和酶解相结合，并利用DNA吸附树脂进行纯化。结果表明它能有效地去除沉积物中的腐殖酸等抑制剂，每克深海沉积物样品可得到DNA约16mg，所得到的DNA分子片段均在23kb左右,可直接应用于各种分子生物学操作。对采集西太平洋“暖池”区两个位点的沉积物样品，通过构建环境样品的微生物16SrDNA克隆文库，采用PCR-RFLP分析、16SrDNA序列测定以及系统发育分析的方...; The microbial diversity and their relationship to the environment in the deep sea sediment from the Pacific were investigated by molecular ecology approaches.
