英文摘要摘要氢酶是微生物氢代谢过程中的关键酶。本研究分离提纯了产酸克雷伯氏菌KlebsiellaoxytocaHP1可溶性氢酶,并对影响该氢酶催化活性的一些理化因素进行测定和分析,为对该氢酶的进一步研究、改造和构建高效产氢的菌株奠定基础。菌体细胞约20g,悬浮于缓冲液中。菌液经超声波破碎和高速离心(75,000×g,40min),上清液过DE-52纤维素柱(1),采用线性梯度(0~0.4mol/LNaCl)洗脱后,收集含可溶性氢酶的洗脱液,再过SephadexG200凝胶柱。其含可溶性氢酶的洗脱组份,经随后的DE-52柱(2)和SephadexG50柱纯化后,可得到纯化约233倍的可溶性氢酶,得率为2...; Abstract Hydrogenases are key enzymes of hydrogen metabolism in many microbes.
