英文摘要以灰绿曲霉(AspergillusglaucusXC9)为原料,经过产酶发酵、离心、硫酸铵逐级盐析、SephadexG-100凝胶层析、PhenylSepharoseFastFlow疏水层析,获得比活力为1747.4U/mg的电泳单一纯β-葡萄糖苷酶(β-1,4-glucosidase,EC3.2.1.21,简称BG)制剂。测定酶分子的亚基分子量约为23.5kDa,全酶分子量采用Sephadex-G100凝胶过滤柱层析法测定为92.5kDa,说明该酶为四聚体。该酶的紫外特征吸收峰在278nm处。在波长280nm的光激发下,该酶的内源荧光发射光谱的发射峰在338nm处。酶催化水杨苷反应的最适温度...; A β-1,4-glucosidase (BG) was purified from fermentation liquor of Aspergillus glaucus by following procedures: ammonium sulfate precipitation, gel filtration on Sephadex G-100, and hydrophobic chromatography on Phenyl Sepharose Fast Flow.
