| 英文摘要 | 南极假丝酵母脂肪酶B（CandidaantarcticlipaseB，CALB）来源于南极假丝酵母，是一种优良的脂肪酶，它对水溶性、非水溶性物质都有很强的催化活性，在有机合成、手性化合物拆分、医药中间体等领域得到广泛应用。然而南极假丝酵母产酶量低，发酵周期长，远不能满足目前的需求。而通过基因改良的米曲霉发酵获得的CALB，其售价约为16000元/千克，不适合工业化应用。本课题主要目的是以毕赤酵母（Pichiapastoris）作为表达系统，通过基因工程手段提高CALB的表达量，从而降低其工业应用的成本。主要研究内容及其结果如下： 首先，探索野生型CALB基因在毕赤酵母表达的可行性。以南极假丝...; Candida antarctica lipase B is an excellent lipase, which shows strong catalytic activity to hydrophilic and hydrophobic substract and is widely used in the resolution of chiral compounds in organic solvent and synthesis of pharmaceutical intermediates.
