英文摘要	目的探讨氯化三乙基锡(TETC)对体外培养大鼠C6脑胶质瘤细胞增殖和凋亡的影响。方法采用MTT法检测0.5,1.0和2.0μMOl/l TETC对体外培养大鼠C6脑胶质瘤细胞作用24 H和48 H后细胞增殖的影响、HE染色细胞形态学观察0.5,1.0和2.0μMOl/l TETC对体外培养大鼠C6脑胶质瘤细胞作用48 H后细胞增殖和形态学变化。采用透射电镜观察0.5,1.0和2.0μMOl/lTETC作用C6细胞48 H后的超微形态学变化。采用荧光原位末端标记方法检测细胞凋亡。结果MTT法及HE染色细胞形态学观察均显示0.5,1.0和2.0μMOl/l TETC在体外均可抑制C6细胞增殖,C6细胞增殖率存在着剂量依赖性和时间依赖性降低趋势;HE染色和透射电镜可观察到C6细胞凋亡的形态学变化。荧光原位末端标记方法检测到细胞凋亡率存在剂量依赖性上升趋势。结论TETC可抑制体外培养大鼠C6胶质瘤细胞增殖,其机制可能主要与C6细胞凋亡有关。; Objective To study the effects of triethyltin chloride(TETC) on the proliferation and apoptosis of cultured rat C6 glioma cells.Methods MTT assay was performed to evaluate the effects of 0.5,1.0 and 2.0 μmol/L TETC on the proliferation of rat C6 glioma cells for 24 and 48 h.HE staining and electron microscope were employed respectively for determining the proliferation effect and detecting the apoptosis of C6 glioma cells treated by 0.5,1.0 and 2.0 μmol/L TETC for 48 h.Fluorescent TUNEL assay was used to measure the percentage of apoptosis in C6 glioma cells treated by 0.5,1.0 and 2.0 μmol/L TETC for 48 h.Results 0.5,1.0 and 2.0 μmol/L TETC could inhibit the proliferation of cultured C6 glioma cells in vitro performed by both MTT assay and HE staining,and the proliferation rate was decreased in a dose-dependent and time-dependent manner.Apoptotic changes were seen in part of TETC treated C6 glioma cells stained with HE under light microscope or in part of TETC treated C6 glioma cells observed by electron microscope.The apoptosis rate,measured by fluorescent TUNEL assay,was increased in a dose-dependent manner in TETC treated C6 glioma cells.Conclusions TETC inhibits the proliferation of C6 glioma cells in vitro,the mechanism of which may be mainly involved in apoptosis of C6 glioma cells.; 国家自然科学基金资助(30070645)
