A Novel Method for Determination of Pepsin Based on Fluorescence Recovery of Aluminium Tetrasulphonated Phthalocyanine-Bovine Serum Albumin Complex Used as Substrate

英文摘要It was found that the fluorescence of aluminium tetrasulphonated phthalocyanine(AlS(4)Pc), a red-region fluorescence probe, was extremely quenched by bovine serum albumin (BSA) in acidic media, but recovered dramatically after the addition of pepsin. Based on this phenomenon, a new method was developed for homogeneous determination of pepsin. Under the optimal conditions(pH 2.5, reaction temperature 50 degrees C and reaction time of 1 h), the linear range of assay was 0.04 - 4 mg/L and the detection limit was 20 mu g/L. The constructed method has been used to the analysis of pepsin tablet samples with satisfied results.
