癸酸钠抗性菌株产达托霉素培养条件的响应面优化; Screening of Sodium Caprate Resistance Strains for Daptomycin Production and Medium Optimization by Response Surface Methodology

英文摘要为提高玫瑰孢链霉菌(STrEPTOMyCES rOSEOSPOruS)nrrl 11379产达托霉素的能力,在实验室已有研究基础上对生产菌株进行癸酸钠抗性筛选,得到癸酸钠抗性株d1000-S3-2-P2000,经验证其能够耐受高癸酸钠浓度.此外,使用因子筛选和响应面设计对发酵培养基进行氨基酸、维生素、微量元素种类的筛选和优化,最终确定培养基组成为(g/l):酵母提取物11,(nH4)2fE(SO4)2.6H2O 0.86,葡萄糖10.7,糊精72,糖蜜7.2,组氨酸1.7,天冬氨酸1.3,l-天冬酰胺0.74,硫辛酸0.007 92,znSO4.7H2O 0.55.利用此培养基进行摇瓶和发酵罐分批发酵培养得到达托霉素产量分别为42.38和411Mg/l,分别是优化前的2.52倍和1.46倍.; In order to improve the production of daptomycin by Streptomyces roseosporus NRRL 11379,sodium caprate resistance screening method was applied to obtain high sodium caprate resistance strains based on original research.A mutant strain D1000-S3-2-P2000 was achieved and had been proved to be with tolerance in high sodium caprate concentraion.Besides,amino acids,trace elements and vitamins of the medium were screened and further optimized by response surface methodology(RSM).The compositions of the optimized medium were as follows(g/L):yeast extract 11,(NH4)2Fe(SO4)2·6H2O 0.86,glucose 10.7,dextrin 72,molasses 7.2,histidine 1.7,aspartic acid 1.3,L-asparagine 0.74,lipoic acid 0.007 92,ZnSO4·7H2O 0.55.The daptomycin production of 42.38 mg/L(flask) and 411 mg/L(2.5 L-batch) were achieved,which were 2.52 and 1.46-fold of those using original conditions.; 福建省自然科学基金项目(2011J01058); 福建省产业技术开发项目(闽发改高技[2010]299号); 中央高校基本科研业务费专项(2011121017)
