| 英文摘要 | 采用PCr-dnA测序技术测定、分析了闽—粤东族、岱衢族大黄鱼群体47个个体的线粒体dnA控制区基因序列.结果表明:所获序列长度为786--799 bP,具32个碱基替换位点和41个插入/缺失位点;控制区碱基组成中T、C、A、g含量分别为31.9%、15.8%、29.3%和23.0%,平均转换颠换比(r)5.5;共有10个单倍型,闽—粤东族8个和岱衢族3个,仅HAP03为共享单倍型;所有个体的平均单倍型多样性(Hd)为0.470,核苷酸多样性(PI)为0.006 50;2个群体的遗传距离(d)为0.007 13,遗传分化指数(fST)为0.181 6,基因流(nM)为1.13.群体遗传分析结果表明,闽—粤东族、岱衢族大黄鱼野生群体遗传变异处于较低水平,闽—粤东族大黄鱼的遗传多样性水平稍高于岱衢族,群体间的遗传分化不明显.; The mitochondrial DNA(mtDNA)control regions of Pseudosciaena crocea sampled from Min-yue and Dai-qu Stock were amplified by PCR and sequencing.A 786--799 bp fragment of control region in mtDNA from 47 individuals of P.crocea was determined,there were 32 polymorphic sites accounting for 4.01% of total analyzed sites and 41 sites with gaps/missing.The base amounts of T、C、A and G were 31.9%,15.8%,29.3% and 23.0%,respectively.The mean ratio of transition to transversion in this study was 5.5.10 haplotypes,8 in Min-yue Stock and 3 in Dai-qu Stock,were defined,in which only one,i.e.Hap03 was shared by the two stocks.The average number of haplotype diversity(Hd) and nucleotide diversity(Pi) were 0.470 and 0.006 50,respectively.The genetic distance(D) and the index of gene differentiation(Fst) were 0.007 13 and 0.181 60,whereas the gene flow(Nm) was 1.13.The results indicated that the genetic diversity of wild P.crocea in Min-yue and Dai-qu Stock was quite low.The genetic diversity in Min-yue Stock seemed to be greater than that in the Dai-qu Stock,but no significant difference in both.; 国家863计划(2006AA10A405);国际科技合作项目(2006DFA32960) |
