中文摘要以茶树菇为原料,通过单因素试验和正交试验,研究了料水比(W/V)、浸提时间、沉淀的乙醇浓度和浸提温度4个因素对茶树菇多糖提取得率的影响;探讨了茶树菇多糖中蛋白质的脱除技术,同时采用生物化学法测定了茶树菇多糖对活性氧自由基O2-·、·OH和H2O2的清除能力。研究结果表明:提取茶树菇多糖的最佳工艺条件为:沉淀的乙醇浓度80%、料水比1∶20、浸提时间180min、浸提温度70℃。经红外光谱和紫外光谱分析,茶树菇多糖主要是吡喃多糖,无蛋白质特征吸收峰。茶树菇多糖具有较强的清除活性氧自由基的能力,当茶树菇多糖质量浓度达0.3mg/mL时,对O2-·、·OH和H2O2的清除率分别为41.72%、31.56%和55.81%;当茶树菇多糖质量浓度增大时,对O2-·、·OH和H2O2的清除率增加,且两者呈一定的量效关系。; Using Agrocybe aegerita as raw material,the effects of ratio of tea to water(W/V),extraction time, concentration of ethanol and extraction temperature on the yield of Agrocybe aegerita polysaccharides were studied by single factor test and orthogonal test,and the Technologies of removing protein from the Agrocybe aegerita polysaccharides(OTPS)were investigated,and scavenging effects of Agrocybe aegerita polysaccharides on active oxygen free radicals were detected by biochemical method. The results showed that concentration of ethanol had significant effect on the yield of polysaccharides,and time of extraction,ratio of tea to water,concentration of ethanol,temperature of extraction had no significant effect. The experimental analysis showed that the optimum technological parameters were:extraction 180 min at 70 ℃ with 1∶22 ratio of tea to water,and 80% of concentration of ethanol. The infrared spectrum and ultraviolet spectrum analysis showed that a majority of Agrocybe aegerita polysaccharides is pyranopolysaccharide,and there is no the typical absorption of protein in it. It was found that the Agrocybe aegerita polysaccharides had a strong scavenging capacity for active oxygen free radicals. When the concentration of Agrocybe aegerita polysaccharides were at 0.3 mg/mL,its scavenging rates to superoxide anion free radical(O2-·),hydroxyl radical(·OH)and H2O2 were 41.72%、31.56% and 55.81%,respectively.
