中文摘要用Fe3+作为筛选剂选择性分离虾青素高产菌株,实验结果表明,海洋红酵母的菌落形成率随平板培养基中Fe3+浓度的增加而降低,当Fe3+浓度为5 mmol/L时,海洋红酵母的菌落形成率降至0;海洋红酵母的虾青素体积产率和细胞产率随摇瓶培养基中Fe3+浓度的增加而降低;在含有5 mmol/L Fe3+的平板培养基中分离甲基磺酸乙酯(EMS)诱变得到的海洋红酵母,大约有31%的菌落虾青素体积产率高于出发菌株.在海洋红酵母的分离培养基中加入较高浓度的Fe3+可以淘汰低产突变株及野生型菌株,可以提高海洋红酵母虾青素高产菌株的筛选效率.; A novel method for selective isolation astaxanthin high-yield mutants of Phaffia rhodozyma with Fe3+ was developed.As shown by the results,increasing the concentration of Fe3+ caused the decrease of colony forming rate,and the colony could grow on the plate containing less than 5 mmol/L of Fe3+.Meanwhile,astaxanthin volumetric yield and specific yield decreased along with the increasing concentration of Fe3+.After Phaffia rhodozyma population mutagenized with EMS,the yeast was separated on selective medium including 5 mmol /L of Fe3+.About 31 % of the colonies were detected which showed higher astaxanthin volumetric yield than the original strain.Thus,the developed method could sife cells of astaxanthin low yield mutants and original strain and increase the efficiency of isolating astaxanthin high-yield mutant of Phaffia rhodozyma.; 【作者单位】集美大学生物工程学院; 厦门市食品生物工程技术研究中心;【作者英文名】LI Li-jun1,2,XIAO An-feng1,2,HUANG Zhi-ying1,CAI Hui-nong1,2,NI Hui1,2(1.School of Biotechnology Engineering,Jimei University,Xiamen 361021,China;2.Food and Bio-engineering Technology Research Center of Xiamen,Xiamen 361021,China)
