中文摘要目的为了降低荧光显微图像的噪声和离焦平面的信息,提高图像清晰度,并评价去卷积成像方法的性能,本文进行显微荧光去卷积成像评价方法的研究。方法首先,将测得的系统点扩散函数作用于共聚焦显微镜采集的endoG-GFP标记的人乳腺癌细胞MCF-7图像,得到模糊图像,然后采用Richardson-Lucy(R-L)算法对模糊的荧光图像进行去卷积运算,并对去卷积成像方法的性能进行了分析和评估。结果显微荧光去卷积成像方法可有效去除焦平面外荧光信息,显著提高荧光图像的清晰度。结论显微荧光去卷积算法结合有效的评价方法在未来有望用于微流控细胞培养的成像,对活体细胞和组织的三维结构的研究有一定的参考价值。; Objective In order to reduce the noise and out-of-focus fluorescence information of fluorescence microscopy images,and to evaluate the performance of deconvolution imaging method,this paper proposed a method to evaluate the image quality of the deconvolution algorithms for fuorescence images.Methods In this paper,the fluorescence MCF-7 images obtained from a confocal laser-scanning microscope system were convoluted by the point spread function(PSF) to get the blurred fluorescence images that were treated as the convolution procedures of the original fluorescence light sources with the PSF of the imaging system.The Richardson-Lucy(R- L) algorithm was applied to the deconvolution methods of the blurred images.Results The algorithm could solve the convolution model of the fluorescence microscope imaging system and reduce the fluorescence light from out-of-focus planes.The best result of current experiments nearly approached the image quality of the CLSM system.Conclusions The R-L deconvolution algorithm combined with effective evaluation methods is expected to be applied to microfluidic cell culture imaging,and have a certain reference value to the study on the three-dimensional structure of living cells and tissues.
