| 中文摘要 | 目的:利用大鼠离体肺再灌注模型,观察特异性磷酸二酯酶抑制剂米力农在器官摘取及保存期对大鼠供体肺的保护作用。方法:24只SD大鼠随机分成对照组和实验组,每组12只。对照组为常规保存组,低钾右旋糖酐液(LPD)灌洗后摘取心肺器官,4℃恒温冰箱中保存12h后取出,以5mL·min~(-1)流量再灌注60 min;实验组供体肺灌洗液及保存液中含有666μg·L~(-1)米力农,余同对照组。结果:实验组供体肺的换气功能、肺血流动力学指标、湿重干重比及超微结构均显著优于对照组(P<0.05),再灌注后肺组织丙二醛(MDA),髓过氧化物酶(MPO)和白细胞介素-8(IL-8)含量均显著低于对照组(P<0.05)。结论:使用特异性磷酸二酯酶抑制剂米力农处理供体肺能够减轻缺血再灌注损伤,明显改善移植后早期供体肺功能。; Objective:To study the protective effect of milrinone,a specific phosphodiesterase inhibitor,on graft lung function during procurement and cold preservation.Methods:Twenty-four SD rats were randomly divided into 2 groups.The lungs were flushed and preserved for 12 h in 4℃low po- tassium dextran (LPD) solution,and then reperfused with blood (5 mL·min~(-1)).The lung preservation and reperfusion were performed in the absence (control) or presence of milrinone (666μg·L~(-1) or 20μg/30 mL).Results:The gas exchange,uhrastructure,wet/dry weight ratio and hemodynamics were superior in lungs treated with milrinone.The contents of MDA,MPO and interlukin-8 in pulmonary tissue after reperfusion were significantly lower in milrinone group than in control group.Conclusions:The spe- cific phosphodiesterase inhibitor milrinone provides protection from ischemia-reperfusion injury in an in vitro model of lung transplantation during cold preservation. |
