中文摘要游离脂肪酶催化法以其酶催化剂生产成本较低,反应速率较快的优点,成为生物柴油制备的新的关注方向。游离脂肪酶NS81006可以有效催化大豆油甲醇解制备生物柴油,但其催化活性在反应后期下降,甲醇可能对NS81006催化活性及回用稳定性造成影响。研究了甲醇对NS81006酶活的影响以及不同甲醇添加策略对NS81006催化效果的影响。研究结果表明,反应体系中过多甲醇的存在对NS81006酶活产生负面影响;通过减少甲醇添加量和缩短甲醇与NS81006作用时间可有效降低酶活损失,提高NS81006回用稳定性;在优化策略下,NS81006连续使用6个批次,生物柴油得率保持90%以上。; Due to its advantage of low cost and rapid reaction rate,soluble lipase catalysis has been considered as a promising way to prepare bio-diesel.Soluble lipase NS81006 could effectively catalyze the methanolysis of soybean oil for bio-diesel production;however,in the later stage of the reaction,the catalytic activity of NS81006 declined,which might be due to the effect of methanol.Effect of methanol on catalytic activity of NS81006 and the effect of different loading methods of methanol on catalytic efficiency were studied.The results indicated that the presence of too much methanol in the reaction medium had a negative effect on lipase activity.Reducing the loading amount of methanol and shortening the interaction time between methanol and NS81006 could effectively decrease the loss of lipase activity and improve its reusing stability.Under the optimal strategy,NS81006 could be reused for 6 batches with the yield of bio-diesel(FAME) at 90% or above.
