英文摘要 | Photothermal therapy (PTT) of cancer has drawn great attention owing to its advantages such as minimal invasion, fewer complications and reduced side effects for normal tissues compared to conventional therapies. Here, we developed one kind of step-by-step responsive nanoparticles (S-S-R NPs) as a smart theranostic platform for noninvasive fluorescence imaging and PTT of cancer. Firstly, indocyanine green (ICG) was mixed to poly(cystamine bisacrylamide spermine) [Poly(CBA-SP)] to form redox-responsive nanoparticles, then the nanoparticles were added to a type of charge-conversion polymer poly-(ethylene glycol)-b-poly(l-lysine)-g-(1,2-dicarboxylic-cyclohexene anhydride) [PEG-b-PLL(DCA)] in water to form the S-S-R NPs which were almost spherical in shape. In the circulation system (pH 7.4), S-S-R NPs would minimize nonspecific interactions with serum components. While under slightly acid pH conditions (pH 6.5), zeta-potentials of the nanoparticles became positively charged, resulting in the formation of shedded nanoparticles. The near-infrared (NIR) images of S-S-R NPs demonstrated that the redox-responsive nanoparticles disassembled and the fluorescence recovered because of the reductive environment in the cytoplasm, which was called the second step responsiveness of the S-S-R NPs. In vitro cellular uptake and PTT efficiency of S-S-R NPs were evaluated usingMCF-7 cells. The results showed that charge conversion could enhance cell endocytosis of S-S-R NPs under acid environment and when irradiated with laser, the viability of cells decreased rapidly. These results indicated that the S-S-R NPs could be a promising theranostic system for cancer imaging and PTT. |
