Remote and noninvasive NIR-triggered DOX release from DOX/ICG-loaded temperature-responsive nanoparticles to promote anti-tumor effect

英文摘要Smart nanoparticles (NPs) that respond to external stimulations, including light, hyperthermia, reduction/oxidation and ultrasound have been developing to achieve optimal drug release in tumor tissues. However, applying these smart NPs to attain high antitumor performance is hampered by limited drug carriers and inefficient spatiotemporal control. In this study, we report a noninvasive NIR-driven, temperature-sensitive nanoparticles (DI-NPs) co-encapsulating doxorubicin (DOX) and indocyanine green (ICG). This theranostic delivery system applies thermoresponsive lipid to controllably release the drug, utilizes the fluorescence (FL) of DOX and ICG to real-time trace the distribution of NPs, and employs DOX/ICG to treat cancer by chemo/photothermal therapy. DI-NPs exhibit uniform size distribution, excellent FL/size stability, enhanced response to temperature, and increased drug release through laser irradiation. DI-NPs also show long retention time, spatiotemporally-controlled release of DOX in tumor tissues, and enhanced ICG and DOX accumulation. The thermo-sensitive DI-NPs manifest a superior efficiency to promote apoptosis of tumor cells, and completely suppress tumor growth without side-effect. This study may provide a new generation of smart strategy for combinatorial cancer therapy.
