Multifunctional theranostic nanoparticles for dual-modality imaging and magnetic targeted photodynamic therapy

英文摘要Photodynamic therapy (PDT) is an effective treatment for cancer with minimal nonspecific damage to normal tissues. To improve the photodynamic therapeutic efficiency, we loaded superparamagnetic Fe3O4 NPs into the core of redoxresponsive Ce6-conjugated dextran nanoparticles (DSSCe6 NPs), obtaining a multifunctional theranostic system (DSSCe6@Fe3O4 NPs) for dual-modality imaging and magnetic targeted PDT. DSSCe6@Fe3O4 NPs were almost spherical in shape and T2-weighted MR imaging revealed a concentrationdependent darkening effect with the transverse relaxivity coefficient (r2) measured to be 172.3 s−1 mM−1, confirming that it is a good contrast agent in T2-weighted MR images. This theranostic system also exhibits significantly accelerated cellular uptake under magnetic field, and thus offers greatly improved in vitro photodynamic cancer cell killing efficiency. These results indicated that the DSSCe6@Fe3O4 NPs could be a promising theranostic system for cancer imaging and photodynamic therapy.
