Improved optrode array design and neural/tissue interface for optogenetics

英文摘要Objective: Optogenetics is a technology that combines optical control and genetic targeting using cell-type specific and optically-sensitive proteins for the precise manipulation of neuronal functions. This technology has been successfully used to understand the mechanisms of neuropsychiatric diseases through the precise spatial and temporal control of specific groups of neurons in a neural circuitry. However, it remains a great challenge to integrate optogenetic modulation with electrophysiological and behavioral read out methods as a means to explore the causal, temporally precise, and behaviorally relevant interactions of neurons in the specific circuits of freely behaving animals. Therefore, the aim of this study is to develop a novel optrode array for long term applications in awaken rodents. Methods: We have designed and fabricated a high spatial resolution, multi-channel, and drivable optrode array for optical stimulating and recording in different brain regions of a rodent model at one time. Meanwhile, we have also developed optimal modification methods to build a low impedance and tissue friendly optrode/neural interface based on electro-active and bio-active materials, such as conducting polymers, hydrogels, neurotrophins and drugs. Results: According to the requirements in the application of implantable optrode arrays, evaluation processes and characterization of the optrode arrays, both in vitro and in vivo, were exploited and established. The investigation includes the electrochemical properties and biocompatibility of the optrode/tissue interface, and the long-term stability of the electrophisiological recording signals in freely-behaving animals. Meanwhile, some other devices based on the technique of optogenetics and electrophysiology were developed, which were used to investigate the complex neural circuit processing mechanisms at the functional level. Conclusion: This work will provide theoretic and technical support for the development of a new generation of optrode arrays, and also for the development of optogenetics. Moreover, it will offer significant advantages and show great potential in both understanding the mechanisms of neuropsychiatric diseases such as Parkinson’s disease, anxiety, schizophrenia,epilepsy, depression, and social dysfunction and in developing new therapeutic strategies.
