中文摘要目的探讨建立树鼩AS模型中NPC1L1(Niemann-Pick C1-Like1)基因在小肠和肝中mRNA的表达差异.即树鼩脂质代谢和动脉粥样硬化的关系.方法通过半定量RT-PCR分别检测树鼩不同膳食组间小肠、肝组织中的NPC1L1 mRNA表达差异.结果高脂饲料组树鼩肠组织中基因NPC1L1 mRNA的表达(3910±262)μmol/L,与正常饲料组比较(P〈0.05),表达水平显著高于正常饲料组.结论树鼩消化道组织NPC1L1表达增加与脂肪代谢小肠组织中NPC1L1的高表达有关,NPC1L1可能参与了血脂紊乱的病理生理过程,NPC1L1与促成动脉粥样硬化的发生机制有待进一步研究.
英文摘要Objective To investigate the expression differences of the Niemann-Pick C1-Like 1(NPC1L1) gene mRNA in the small intestine and liver of atherosclerotic tree shrews.That is,the relationship of tree shrews lipid metabolism and atherosclerosis.Methods Measured the expression differences of the NPC1L1 gene mRNA in the small intestine and liver of atherosclerotic tree shrews among different dietary groups by means of semi-quantitative RT-PCR detection,respectively.Results The NPC1L1 mRNA expression in intestinal tissue of tree shrews fed a high-fat diet was(3910±262) nmal/L,and the expression level was significantly above that of normal forage(P 0.01).Conclusion The increase of NPC1L1 expression in gastrointestinal tissue and fat metabolism of tree shrews were related to the high expression of NPC1L1 in the small intestine.NPC1L1 might participate in the pathophysiological process of dyslipidemia.The mechanism of NPC1L1 in the occurrence of atherosclerosis is needed to be further research.
