中文摘要生物气溶胶是大气与其中的生物微粒所组成的气体分散体系。生物气溶胶浓度和组分直接影响人类的健康,以其为施放形式的生物战剂是生物恐怖袭击的主要方式。为了快速应对与处置有害生物气溶胶,迫切需要一种能对其进行预警的实时监测的技术与设备。 本文概括了光学生物气溶胶检测的几种手段,包括粒径测量、形状识别与荧光检测等;重点分析了本征荧光检测原理和构成生物气溶胶颗粒的几种有机分子及其光学特性。 针对生物气溶胶粒子的本征荧光特性,本文基于已有技术展开了实时生物气溶胶监测技术的研究,研制了一套280nm波长紫外光诱导荧光检测系统,实现了对富集到的环境空气中的生物气溶胶粒子的本征荧光探测与预判别。利用本荧光检测系统对色氨酸、NADH和牛血清蛋白进行了荧光检测与光谱分析,讨论了本系统对于紫外光诱导荧光检测与对生物气溶胶粒子进行判别的可行性。 为了验证研制的紫外光诱导荧光检测系统的可靠性,提出了一套针对该实时监测系统的标定方法与装置,以色氨酸为标定物,借助光学粒子计数器,对该荧光检测系统进行标定。标定实验结果表明,本系统对于生物气溶胶荧光检测具有良好的线性响应特性和稳定性。
英文摘要Biological aerosol is a dispersion system consisted of atmosphere and its biological particles. The concentration and composition of bio-aerosol have a direct impact on human health, and biological warfare agent aerosol is the main way of bioterrorist attack distributed as the form of bio-aerosol. In order to quickly respond to and dispose the hazardous aerosols, there is an urgent need of a real-time monitoring equipment technology and equipment for the biological aerosol warning. This article summarizes several means of optical bio-aerosol detection including measurement of particle size, shape recognition and fluorescence detection. Fluorescence detection principle and several sorts of organic molecules containing in biological aerosol particles and their optical properties are emphatically introduced. According to intrinsic fluorescence characteristics of the biological aerosol particles, a research on real-time bio-aerosol monitoring techniques was carried out based on existing technologies, and a UV light induced fluorescence detection system was proposed and developed. Such system could implement the detection and pre discrimination of biological aerosol particles in ambient air. By fluorescence detection and spectral analysis of tryptophan, NADH, and bovine serum albumin using this detection system, the feasibility of this system for UV-induced fluorescence detection and pre discrimination of biological aerosol particles are discussed. In order to verify the reliability and stability of the 280mnm UV-induced fluorescence detection system, a calibration method and device is designed with tryptophan as the target, and optical particle couner as the subsidiary. Calibration experiment results show that the design of the calibration system has good linearity, indicating a high degree of linear responsibility and stability of the intrinsic fluorescence detection system.
